Enblend used GPU acceleration for a while (add --gpu to the command line). Although the upcoming release will also accelerate using OpenMP and then you'll have a choice either/or which will depend on how many and how fast your CPUs and your GPUs are.

Hugin since version 0.8.0 uses the GPU for fast preview and you can interact in real time with the panorama (drag images around, pan/tilt/roll/crop the output panorama, etc.). Since version 2009.2 it has also GPU acceleration of the stitching process. Still experimental, may not work for you. It's a checkbox in the preferences. Some users have reported a significant speed increase.
