You did not miss anything and have very good questions.? I asked the same
questions!? Initially I wanted to set up a genetic test to determine the level
of homogenization.? A simple test using a visible genetic marker or a more
involved molecular analysis.? Then, my wife, being the smarter one...suggested
using a simple dye.? At first, I was skeptical there was?significant mixing
taking place in the chamber, but two things changed that view.? One, there is
mechanical mixing that occurs during the semen collection process.? The plunger
is designed so that rotation provides a mechanical mixing inside the chamber.?
The pictures shown do not account for the mechanical mixing as?the pictures
indicate mixing after the semen collection was completed.? To determine if the
movement of the dye was simply due to the penetrating property of the dye, I
replicated the procedure using different diameters of tubing and found the
movement of the dye was not proportional to the diameter of the tubing/chamber
used.? Using standard 100ul semen storage tubes filled with semen the dye does
not penetrate more than 1-2mm, even after several days.?

Thanks for the post and a Happy New year back!
What I did not understand exactly was how you arrived at the conclusion
that there was in fact a homogenized mixture of semen 9,5 hrs after
having added some dye. Wasn't the penetrating property of the added dye
the reason for the spreading of the dye through the drawn up semen? And
if that is so then a homogenizing process of semen may not have taken
place or at least not exactly in line with the shown spreading of the
dye through the semen.
What mechanical part of the syringe is according to you promoting the
homogenizing process?
Did I miss something?
